Bacillus subtilis is a kind of Gram-positive bacteria with good characteristics and has been widely used in the production of heterologous proteins. Red / ET recombination can use phage protein pair (RecE / RecT or Reda / Redb) mediated homologous recombination to carry out DNA engineering in Bacillus subtilis. The core step of Red / ET recombination is the crossover step between the targeting construct containing homology arms and the target. The target can be the locus of Bacillus subtilis. By designing a homologous fusion fragment of the target gene, clone it to suicide. In the vector, the suicide vector is then transformed into the target bacteria. The insertion mutants were selected by antibiotic screening. Under the second round of negative selection pressure, only mutations that include the second homologous recombination and suicide plasmid loss can be spared. Through PCR screening and sequencing, researchers can obtain mutants of this strain. The specific workflow includes homology arm design and suicide plasmid construction, recombination, selection/screening, verification, etc.

CRISPR technology is derived from the immune system in bacteria and archaea. It is an effective genome-scale editing tool. It has completely changed the traditional genetic engineering methods and promoted strain engineering as never before. It enables Bacillus subtilis to perform genetic manipulations quickly and reliably. Two components are needed to work: a guide RNA (gRNA), such as under the RNA polymerase III promoter, and a DNA endonuclease fused with a nuclear localization tag, of which Cas9 is the most commonly used. The project process includes sgRNA design and construction, transformation, selection/screening, and verification.
